A once-a-day controlled release formulation of a water soluble drug is
described which includes:
(a) from 20 to 50% by weight of enteric polymeric membrane coated pellets
comprising a polymer membrane coated core which comprises a biologically
inert core which is coated with a first layer which consists essentially
of a water soluble drug and a binder; and a second layer which comprises a
membrane comprising a pH dependent polymeric material; and
(b) from 50% to 80% by weight of delayed pulse polymeric membrane coated
pellets comprising a polymeric membrane coated core which comprises a
biologically inert core which is coated with a first layer which consists
essentially of a water soluble drug and a binder and a second layer which
comprises a polymeric membrane and a alkaline earth metal stearate which
will substantially maintain its integrity in the varying pH conditions of
the gastrointestinal tract but is permeable to said water soluble drug;
and
(c) a unit dose containment system.
On décrit une formulation de dégagement commandée parjour d'une drogue hydrosoluble qui inclut : (a) de 20 à 50% du poids de la membrane polymère entérique a enduit des granules comportant un noyau enduit par membrane de polymère qui comporte un noyau biologiquement inerte qui est enduit d'une première couche qui consiste essentiellement en une drogue hydrosoluble et une reliure ; et une deuxième couche qui comporte une membrane comportant un matériel polymère dépendant de pH ; et (b) de 50% à 80% en poids de retardé impulsion polymère membrane a enduit granule comportant polymère membrane enduit noyau qui comporte biologiquement inerte noyau qui est enduit de premier couche qui consiste essentiellement en une drogue hydrosoluble et une reliure et une deuxième couche qui comporte une membrane polymère et un stéarate alcalinoterreux qui maintiendront sensiblement son intégrité en conditions variables de pH de l'appareil gastro-intestinal mais est perméable à ladite drogue hydrosoluble ; et (c) un système de retenue de dose d'unité.
