Compounds and methods are provided for use in purification of CD34.sup.+ cells and specific surface antigens thereof. The present invention discloses methods for releasing CD34.sup.+ cells, as well as compounds having a carbohydrate epitope of the CD34 surface antigen, from an affinity matrix, using carbohydrates having the structure: Neu5Ac.alpha.2-3Gal.beta.1-4(X) wherein (X) is GlcNAc, or a monosaccharide or a cyclohexane derivative that is structurally similar to GlcNAc.
