The present invention relates to Glutathione S-transferase (GST)/Reduced
Glutathione (GSH) as a means for the in-vivo release of a drug that has
been conjugated to specific electrophilic moieties via a sulfonamide bond.
The drug may be an anticancer agent (or one with other therapeutic
properties) carrying a free --NH-- which has been derivatized by the
attachment of an electrophile containing a moiety, such as p-CN-- or
p-NO.sub.2 -pyridinylsulfonyl groups, or p-NO.sub.2 - or 2,4
dinitrophenylsulfonyl groups, or suitable derivatives thereof, to make a
prodrug. Optionally, the sulfonamide moiety may have attached to it a
targeting molecule. The present invention also provides Glutathione
S-transferase (GST)/Reduced Glutathione (GSH) as a means for the release
of a protected amino derivative that has been conjugated to specific
electrophilic moieties via a sulfonamide bond. The precursor is a
synthetic intermediate carrying a free --NH-- which has been derivatized
by the attachment of an electrophile via a sulfonamide bond.
Die anwesende Erfindung bezieht auf Glutathion S-Transferase (GST)/Reduced Glutathion (GSH) als Mittel für die in vivo Freisetzung von einer Droge, die zu den spezifischen elektrophilen Hälften über eine Sulfonamidbindung konjugiert worden ist. Die Droge kann ein krebsbekämpfendes Mittel (oder eins mit anderen therapeutischen Eigenschaften) sein ein freies --NH tragend -- das durch das Zubehör von einem electrophile derivatisiert worden ist, eine Hälfte, wie p-CN -- oder p-NO.sub.2 enthalten - pyridinylsulfonyl Gruppen oder p-NO.sub.2 - oder 2.4 dinitrophenylsulfonyl Gruppen oder verwendbare Ableitungen davon, um einen Prodrug zu bilden. Beliebig kann die Sulfonamidhälfte zu ihr ein zielendes Molekül angebracht haben. Die anwesende Erfindung liefert auch Glutathion S-Transferase (GST)/Reduced Glutathion (GSH) als Mittel für die Freisetzung von einem geschützten Aminoderivat, das zu den spezifischen elektrophilen Hälften über eine Sulfonamidbindung konjugiert worden ist. Der Vorläufer ist ein synthetischer Vermittler, der ein freies --NH trägt -- das durch das Zubehör von einem electrophile über eine Sulfonamidbindung derivatisiert worden ist.
