Disclosed are methods for allele discrimination involving the use of
rolling circle amplification (RCA) coupled with primer extension and
utilizing exonuclease deficient polymerases to distinguish matched and
unmated single nucleotide sites, such as in the case of a single
nucleotide polymorphism (SNP).
