Robust stain detection and quantification for histological specimens based on a physical model for stain absorption

   
   

A physics based model of the absorption of light by histological stains used to measure the amount of one or more stains at locations within tissue is disclosed. The subsequent analysis results in several improvements in the detection of tissue on a slide, improvements to autofocus algorithms so focusing during image acquisition is confined to tissue, improvements to image segmentation and identification of tissued and its features, improvements to the identification of stain where multiple stains are used, and improvements to the quantification of the extent of staining. The invention relates to the application of these improvements to stain detection and quantification to provide for objective comparison between tissues and closer correlation between the presentations of such features and concurrent patterns of gene or protein expression.

Una fisica ha basato il modello dell'assorbimento di luce dalle macchie istologiche usate per misurare la quantità di una o più macchie alle posizioni all'interno del tessuto è rilevata. L'analisi successiva provoca parecchi miglioramenti nella rilevazione del tessuto su uno scorrevole, miglioramenti alle procedure di autofocus in modo da mettere a fuoco durante l'aquisizione di immagine è limitata al tessuto, miglioramenti a segmentazione di immagine e l'identificazione di tissued e le relative caratteristiche, miglioramenti all'identificazione di macchia in cui le macchie multiple sono usate e miglioramenti alla quantificazione del limite di macchiatura. L'invenzione riguarda l'applicazione di questi miglioramenti a rilevazione ed a quantificazione della macchia per prevedere il confronto obiettivo fra i tessuti e la correlazione più vicina fra le presentazioni di tali caratteristiche ed i modelli simultanei dell'espressione della proteina o del gene.
