The invention provides compounds of Formula (I), 1
stereoisomers thereof, or pharmaceutically acceptable salts of said
compounds or stereoisomers wherein R1 and R2 are as defined herein as
well as compositions comprising the same, processes for making the same,
and methods of using the same to treat a variety of diseases, including,
those requiring interaction with metalloproteases, and more specifically
with macrophage metalloelastase (MMP-12), and for the prevention and
treatment of respiratory pathologies such as chronic obstructive
bronchopneumopathy (COPD), emphysema, chronic bronchitis, chronic
pulmonary inflammation, asthma, cystic fibrosis, acute respiratory
distress syndrome (ARDS), respiratory allergies including allergic
rhinitis, and also diseases associated with the production of TNF.alpha.
including severe fibrotic pulmonary disease, pulmonary sarcoidosis and
silicosis. The compounds of the present invention also show inhibitory
activity on metalloprotease-13 (MMP-13), making them useful for the
treatment of pathologies involving this enzyme, such as cancer,
osteoporosis, osteoarthritis, arthritis, rheumatoid arthritis,
atherosclerosis, multiple sclerosis and cardiac insufficiency.
L'invenzione fornisce i residui della formula (i), stereoisomeri 1 di ciò, o farmaceuticamente sali accettabili dei residui o stereoisomeri in cui R1 e R2 sono come definiti qui così come le composizioni che contengono lo stesso, processi detti per fare lo stesso ed i metodi del usando lo stesso per trattare una varietà di malattie, includente, di quelle che richiedono l'interazione con i metalloproteases e specificamente con il metalloelastase del macrofago (MMP-12) e per la prevenzione ed il trattamento delle patologie respiratorie come bronchopneumopathy ostruttivo cronico (COPD), l'enfisema, la bronchite cronica, l'infiammazione polmonare cronica, l'asma, la fibrosi cystic, la sindrome respiratoria acuta di afflizione (ARDS), le allergie respiratorie compreso allergico il rhinitis ed inoltre le malattie si sono associati con la produzione di TNF.alpha. compreso la malattia polmonare fibrotica severa, la sarcoidosi polmonare e la silicosi. I residui di presente invenzione inoltre mostrano l'attività inibitoria su metalloprotease-13 (MMP-13), rendente li utili per il trattamento delle patologie che coinvolgono questo enzima, quali cancro, osteoporosis, la osteoartrite, l'artrite, l'artrite rheumatoid, l'aterosclerosi, la sclerosi a placche e l'insufficienza cardiaca.
