Methods and agents are disclosed for modulating the interaction of .beta.-catenin or .gamma.-catenin with CBP or p300. Agents that increase the binding of CBP to .beta.-catenin are associated with enhancing the .beta.-catenin-related proliferation of adult stem cells, including hematopoietic stem cells, neural stem cells, skin stem cells, and pancreatic stem cells.
