This invention provides a method for rapidly obtaining accurate three-dimensional structure of proteins including large or multi-sub unit proteins, using a combination of NMR analysis of backbone only .sup.13 C, .sup.15 N or .sup.13 C and .sup.15 N isotopically labeled proteins which are optionally also .sup.2 H isotopically labeled in the C.alpha. position protons and residual dipolar coupling measurements in more than one partially aligned state and/or orientation data pertaining to overlapping successive peptide pairs.

Deze uitvinding verstrekt een methode om nauwkeurige driedimensionele structuur van proteïnen met inbegrip van grote of multi-subeenheidsproteïnen snel te verkrijgen, slechts gebruikend een combinatie van NMR analyse van backbone sup.13 C, sup.15 geëtiketteerde proteïnen van N of sup.13 van C en sup.15 van N de isotopisch die naar keuze ook sup.2 H isotopisch dat in de C.alpha. positieprotonen en de overblijvende tweepolige koppelingsmetingen wordt geëtiketteerd in meer dan één gedeeltelijk gericht staat en/of richtlijngegeven betreffende overlappende opeenvolgende peptide paren zijn.
